Aussie Olympian Lisa Weightman broke the record in the women's event, clocking a blitzing 2:26.05 - more than six minutes quicker than the previous record.
Melbourne's weather was initially kind to around 34,000 runners - the most ever - before showers moved in.
Competitors are tackling five distances, including a 3km walk and 21.1km half-marathon, which ended at the MCG.
About 7000 were expected to complete the full course, topping last year's figure of 6200.
"It will be a massive record for an Australian marathon,'' competition director Tim Crosbie said.
"All the hard work is coming to fruition today.
"There's a lot of satisfied but tired people."
The marathon, first held in 1978, this year raises money for the Cerebral Palsy Education Centre.
